Historically, deconvolution was considered unnecessary in confocal microscopy and limited to the enhancement of widefield images. Now, every major microscope manufacturer discusses deconvolution of confocal images as a tool for enhancing resolution of these images. This paradigm shift requires careful attention to the deconvolution methods and associated artifacts. Both linear and nonlinear deconvolution methods will be discussed and the utility of these methods for enhancing image resolution. GPU-acceleration improvements in deconvolution and easy imaging modality selection are now offered by Olympus. These improvements in deconvolution methods and how these methods are evolving in modern microscopy will be discussed.
